چکیده

Since Knox and Grossman demonstrated the excretion by man of the 6-pyridone of N’-methylnicotinamide (hereafter referred to as the pyridone) in appreciable amounts after the ingestion of nicotinamide (2), it became necessary to devise a method for its quantitative estimation in urine. Knox and Grossman isolated the compound from human urine and described its properties, including its ultraviolet absorption spectrum. They found that this metabolite did not yield a fluorescent derivative under the conditions employed for the determination of N1-methylnicotinamide (3); nor could it be determined microbiologically with the use of the customary organisms. The above observation by Knox and Grossman was confirmed that the condensation reaction with acetone in an aqueous alkaline medium which yields a fluorescent product with N1-methylnicotinamide and the pyridone nucleotides (3) did not yield a fluorescent compound with the pyridone. It was soon found, however, that a fluorescent compound could be obtained by agitating the pyridone in acetone solution with KOH in the presence of a minimal amount of water. With the applicability of this reaction to analytical purposes in pure solutions of the pyridone established, it was then necessary to find a method of separation and purification of relatively small amounts of the pyridone present in normal human urine which would yield reasonable recovery and blank values. This proved to be extremely difficult. The usual adsorption and elution techniques either were altogether inapplicable or gave variably low recovery values. The final resolution of this difficulty was found in two steps: the removal of pigments and of other interfering substances in urine with lead

The excretion of N-methyl-2-pyridone-5-carboxamide by man following ingestion of several known or potential precursors.

The development of a rapid method for the determination of N-methyl2-pyridone-5-carboxamide (pyridone) in urine (1) has made it practical to attempt to confirm and extend the studies of Holman and de Lange (2) and others (3-5) concerning the excretion of the pyridone following the administration of various known or possible precursors to man. In the conversion of nicotinic acid to the pyridone ...

Metabolic response of humans to ingestion of nicotinic acid and nicotinamide.

The identification of nicotinamide-N1-oxide as a metabolite in the urine of a schizophrenic patient prompted a study of the relative metabolism of nicotinic acid and nicotinamide in mental patients and healthy volunteers. Metabolites quantified included N1-methyl-2-pyridone-5-carboxamide, N1-methyl-4-pyridone-3-carboxamide, N1-methylnicotinamide, nicotinuric acid, and nicotinamide-N1-oxide. Mor...

A simple method for the determination of N1-methylnicotinamide in urine.

N1-methylnicotinamide (MNA) is found in urine as an assymilation product when administered as nicotinic acid or nicotinamide. MNA was extracted from urine as a pure product and its physicochemical properties reported (4-6). Its maximum adsorption is reported by Hochberg, Melnick, and Oser (4) to lie at 265mƒÊ, E1cm1% 265mƒÊ=258 at pH 9.7. It is stable in urine in the dark for one week, even in ...

Estimation of aldehyde oxidase activity in vivo from conversion ratio of N1-methylnicotinamide to pyridones, and intraspecies variation of the enzyme activity in rats.

The in vivo conversion ratio of N1-methylnicotinamide (NMN) to N1-methyl-2-pyridone-5-carboxamide (2-PY) and N1-methyl-4-pyridone-3-carboxamide (4-PY) as a parameter for the estimation of aldehyde oxidase level in rats was examined. NMN and its pyridones (2-PY and 4-PY) are usually detected in the urine of rats. When we measured the ratio of the amount of pyridones to the total amount of NMN an...

Preparation of 2- and 6-pyridones of N1-methylnicotinamide.

In the course of experiments designed to elucidate the structure of reduced diphosphopyridine nucleotide (DPNH) (12), it became necessary to prepare the 2and 6-pyridones of iV-methylnicotinamidel by oxidation of the latter. Huff (5) has described the preparation of N’-methyl-6-pyridone-3-carboxylic acid by alkaline ferricyanide oxidation of either trigonelline or N’-methylnicotinamide. The acid...
